Immunocytochemistry and protein blotting of caspase-9. Immunocytochemistry was performed 24 h after treatments using antibody against cleaved caspase-9 (green color) and counterstained with DAPI (blue color). Western blots were performed using antibodies against total and cleaved caspase-9 in the cytosolic fraction. Beta-tubulin served as an internal control. Relative band intensities were measured and presented in the bar graph. Cleaved caspase-9 protein band intensity increased from 21.8 ± 1.8 in control to 38.4 ± 1.7 after glutamate incubation and reduced to 27.1 ± 1.1 by selenite treatment.
